Chiefs have been blowing hot and cold since the start of the season and will be looking to redeem themselves after a 2-0 defeat to Cape Town City on Tuesday.
Amakhosi have not won major silverware since 2015 and will be desperate to keep their hopes of ending that spell alive.
The Glamor Boys have won the tournament under current sponsors twice, in 2008 and 2014.
Arthur Zwane’s charges will be without midfielder Yusuf Maart, who will be serving the last of his two-match suspension.
Stellenbosch, meanwhile, had a steady start to the season but lost for the first time in midweek after a 3-0 drubbing by DStv Premiership champions Mamelodi Sundowns.
Steve Barker’s charges are competing for the first time in the tournament after finishing fourth on the log last season and will be looking to cause an upset, especially after completing a league double over Chiefs last campaign.
Kick-off has been delayed and the match will start at 15:30.
